Genetic investigation of swimbladder inflation anomalies in the European sea bass, Dicentrarchus labrax L. ArchiMer
Peruzzi, Stefano; Westgaard, Jon-ivar; Chatain, Beatrice.
Although the actiology of swimbladder inflation anomalies in important aquaculture species such as European sea bass D. labrax is not fully determined, culture conditions are commonly suggested as main contributory factors. Little information is available on whether swimbladder inflation has a genetic basis for its expression too. In this work, 24 full-sibling sea bass families from a 4 dams x 6 sires factorial crossing were reared under communal conditions. The larvae developing normal and abnormal (uninflated or hyper-inflated) swimbladders were genotyped at four microsatellite loci, Labrax-3, Labrax-13, Labrax-17, Labrax29, and allocated to the individual breeders. Out of 273 offspring, 97% could be assigned to a single parental pair. The genotype and...

Genetic variation for carcass quality traits in cultured sea bass (Dicentrarchus labrax) ArchiMer
Saillant, Eric; Dupont-nivet, Mathilde; Sabourault, Marie; Haffray, Pierrick; Laureau, Stanislas; Vidal, Marie-odile; Chatain, Beatrice.
Genetic parameters for carcass quality traits were estimated in 27 families of sea bass (3 dams x 9 sires factorial mating design), raised mixed in the same tanks starting before hatching. O. spring parentage was determined a posteriori using 6 microsatellite loci. Carcass quality traits were recorded at 818 days post fertilization (mean standard length: 32.6 +/- 3.1 cm). Genetic parameters were estimated from the sire half sib variance and covariance components. Heritability of body weight (BW) and carcass processing traits (standardized to body weight) percent head weight (Head%), percent viscera weight (Viscera%) and percent visceral fat weight (VisceFat%) were relatively high ranging from 0.48 +/- 0.15 (Viscera%) to 0.87 +/- 0.23 (Head%); the estimate...

Genetics of European seabass (Dicentrarchus labrax L.) ArchiMer
Chatain, Beatrice; Chavanne, Herve.
The production of sea bass, a major species in Mediterranean fish farming, still relies in many cases on the production of fingerlings issued from wild breeders. Still, genetic improvement is a major key to sustainable development for all optimized forms of animal production. Three types of genetic improvement can be acheived in fish: i) the production of monosex populations. For seabass, monosex female populations would be the objective as females grow 20-30% faster than males. However, sex determination in this species is complex, impacted by both genetic and environmental factors. Hence, production of monosex populations is not yet a routine operation, although in some cases almost 100% female batches have been obtained; ii) chromosome set...

Genotype by environment interactions for growth in European seabass (Dicentrarchus labrax) are large when growth rate rather than weight is considered ArchiMer
Dupont-nivet, Mathilde; Karahan-nomm, Bilge; Vergnet, Alain; Merdy, Olivier; Haffray, Pierrick; Chavanne, Herve; Chatain, Beatrice; Vandeputte, Marc.
Two hundred fifty three full-sib families from 33 males and 23 females of European seabass were produced in a partly factorial mating design. All fish were reared in the same tank for 14 months until reaching mean weight of 35 g. then 7000 of them were individually tagged and weighed, and dispatched to four farms in different locations (France, Israel, Italy and Portugal) representing a wide variety of environmental conditions. Around mean weight of 400 g. 1177 to 1667 fish at each site were weighed. Daily growth coefficient (DGC) was calculated. Pedigrees were successfully redrawn for 99.2% of fish using microsatellite markers. Genetic correlations between sites were high for body weight (>0.80 in all cases but one, i.e., five cases over six), but only...

Haematological and physiological characteristics of diploid and triploid sea bass, Dicentrarchus labrax L. ArchiMer
Peruzzi, Stefano; Varsamos, Stamatis; Chatain, Beatrice; Fauvel, Christian; Menu, Bruno; Falguiere, Jean-claude; Severe, Armelle; Flik, Gert.
The purpose of this study was to determine whether diploid and triploid sea bass differed in terms of main haematological and physiological characteristics. Diploid and triploid fish were produced by sub-optimal pressure treatments and held in communal environments under standard rearing conditions. Total red blood cell count (RBCC), haemoglobin concentration (Hb), hematocrit (Hct), mean cell volume (MCV), mean cellular haemoglobin content (MCH), mean cell haemoglobin concentration (MCHC), plasma metabolites, osmotic pressure, gill Na+/K+-ATPase activity, electrolytes, cortisol, and 3,5,3'-triiodo-L-thyronine (T-3) were measured and compared. Triploidisation in sea bass led to an increase in erythrocyte size (32% in cytoplasm surface area, and 50% in...

Heritabilities and GxE interactions for growth in the European sea bass (Dicentrarchus labrax L.) using a marker-based pedigree ArchiMer
Dupont Nivet, M; Vandeputte, Marc; Vergnet, Alain; Merdy, O; Haffray, P; Chavanne, H; Chatain, Beatrice.
253 full-sib families from 33 males and 23 females of European seabass were produced in a partly factorial mating design. All fish were reared in the same tank during 14 months, then 7000 of them were dispatched in four farms to different locations (France, Israel, Italy, Portugal) representing a wide variety of environmental conditions. Around 400 g mean weight, 1177 to 1667 fish in each site were weighed and length was measured. Condition factor (K) was calculated. Pedigrees were redrawn a posteriori using microsatellites markers: parental origin could be retraced for 99.2% of fish. Due to a high incidence of deformities, the useful sample size was reduced to 491-670 fish per site. Maternal effects were small. Using a simple animal model, heritability...

High diversity of skin-associated bacterial communities of marine fishes is promoted by their high variability among body parts, individuals and species ArchiMer
Chiarello, Marlene; Villeger, Sebastien; Bouvier, Corinne; Bettarel, Yvan; Bouvier, Thierry.
Animal-associated microbiotas form complex communities, which are suspected to play crucial functions for their host fitness. However, the biodiversity of these communities, including their differences between host species and individuals, has been scarcely studied, especially in case of skin-associated communities. In addition, the intraindividual variability (i.e. between body parts) has never been assessed to date. The objective of this study was to characterize skin bacterial communities of two teleostean fish species, namely the European seabass (Dicentrarchus labrax) and gilthead seabream (Sparus aurata), using a high-throughput DNA sequencing method. In order to focus on intrinsic factors of host-associated bacterial community variability,...

Human chorionic gonadotropin induces spermatogenesis and spermiation in 1-year-old European sea bass (Dicentrarchus labrax): Assessment of sperm quality ArchiMer
Schiavone, R; Zilli, L; Vilella, S; Fauvel, Christian.
The aims of the present study were (a) to compare sperm quality (percentage of motile spermatozoa, motility duration, density and fertility after cryopreservation) between precocious and normally maturing male European sea bass Dicentrarchus labrax, (b) to examine the potential of human chorionic gonadotropin (hCG) to increase spermiation in precocious males and (c) to examine the potential of hCG to induce spermatogenesis and spermiation in non-precocious 1-year-old males. One hundred precocious and 100 non-precocious fish were each randomly divided in two groups each: control (precocious saline-treated and non precocious saline-treated) and treated (precocious hCG-treated and non precocious hCG-treated). Treated groups were administered weekly with 1000...

Husbandry stress during early life stages affects the stress response and health status of juvenile sea bass, Dicentrarchus labrax ArchiMer
Varsamos, S; Flik, G; Pepin, Jean-francois; Bonga, S; Breuil, Gilles.
In aquaculture management it is important to establish objective criteria to assess health and welfare of the fish. Here we show that European sea bass (Dicentrarchus labrax) confronted with husbandry-associated stress (tank cleaning, i.e. scrubbing, and water temperature variation) during early life stages show poorer survival and disease resistance as juveniles. We evaluated several parameters for stress (plasma cortisol, glucose and lactate, hydromineral status), growth performance, the immune response (plasma IgM levels) and the effects of a nodavirus challenge. Principal component analysis allowed the establishment of a stress panel including plasma cortisol, osmolality, IgM levels and weight. Sea bass juveniles reared during early life in high and...

Impact of a plant-based diet on behavioural and physiological traits in sea bass (Dicentrarchus labrax) ArchiMer
Benhaim, David; Begout, Marie-laure; Pean, Samuel; Manca, Michael; Prunet, Patrick; Chatain, Beatrice.
Replacing aquaculture feeds based on fisheries-derived resources with plant-based diets could be a relevant strategy to improve the sustainability of aquaculture. Recent studies on sea bass have shown that the total and early replacement of marine products by plant products would have a moderate effect on fish growth and body lipid content. Whether a plant-based diet impacts behavioural and physiological traits possibly linked to fish welfare, is not known, however. Here, we studied the effect of a totally plant-based diet introduced at an early stage of sea bass development on self-feeding behaviour, learning ability in a T-maze and stress biomarkers. We first compared learning processes in self-feeding conditions, between naive fish fed a plant-based...

Impact of aquaculture on the genetic structure of Mediterranean populations of Dicentrarchus ArchiMer
Bahri Sfar, Lilia; Lemaire, Christophe; Chatain, Beatrice; Divanach, Pascal; Ben Hassine, Oum Kalthoum; Bonhomme, Francois.
A phylogeographic analysis based on 6 polymorphic microsatellites was performed on 15 sea bass samples. five from Western Mediterranean, seven from Eastern Mediterranean and three coming from French aquaculture stocks. Among the eastern samples, three did not cluster according to their geographic origin but rather with the occidental group. Furthermore. a somewhat lower allelic diversity was observed within these particular samples, indicating that they originated from a limited number of progenitors of foreign origin. Among the aquaculture stocks, only one revealed a significant reduction of genetic variability, indicating that these stocks are largely outbred and open to wild fishes. The fact that the use of occidental fingerlings to seed broodstocks in...

Improvement of feed intake through supplementation with an attractant mix in European seabass fed plant-protein rich diets ArchiMer
Dias, Jorge; Gomes, Ef; Kaushi, Sj.
The incorporation of an attractant mixture by substitution of an equivalent amount of basal dietary mixture was studied in terms of feed intake and growth performance in European seabass juveniles (mean initial weight : 17 g) fed plant-protein rich diets during 21 days. Three diets had as main protein sources; fish meal, or one of two soy protein concentrates (S and E). To the basal mixture of these last two diets, as well as to a corn-gluten based diet, an attractant amino acid mixture was added at a 2.5% level. Daily feed intake was measured throughout the experimental period. Feed intake and weight gain were highest in seabass fed the fish meal based diet. In those fed the soy protein concentrate diets, the addition of the attractant mixture improved...

In search for indirect criteria to improve feed utilization efficiency in sea bass (Dicentrarchus labrax) Part II: Heritability of weight loss during feed deprivation and weight gain during re-feeding periods ArchiMer
Grima, Laure; Chatain, Beatrice; Ruelle, Francois; Vergnet, Alain; Launay, Amandine; Mambrini, Muriel; Vandeputte, Marc.
Selective breeding to improve residual feed intake (RFI) in sea bass (Dicentrarchus labrax) is a major goal that would optimize economic gain while minimizing the environmental impact of production. Due to the difficulty in accurately measuring individual feed intake, no selective breeding program has yet been started. In a previous study, we identified a criterion phenotypically related to RFI variations: the loss of weight during feed deprivation, FD. Moreover, an additional composite criterion (CC) integrating both FD and weight gain during subsequent re-feeding (RF) was closely related to RFI, even though the relationship was only close to significance (P=0.06). The aim of the present study was to estimate the heritability of these two traits, in order...

Incorporation of dietary fatty acid in European sea bass (Dicentrarchus labrax) - A methodological approach evidencing losses of highly unsaturated fatty acids ArchiMer
Robin, Jean; Skalli, A.
A quantitative approach is presented to evaluate fatty acid incorporation in fish. Fatty acid composition of European sea bass juvenile was studied during an experiment using 6 isoproteic (54%) and isolipidic (18%) diets containing 0.23, 0.56, 0.72, 0.86, 1.01 and 1.86% DM n-3 highly unsaturated fatty acids (n-3 HUFA). Whole body fatty acid compositions were studied at the beginning and after 52 and 81 days feeding, fatty acid profiles evolved during time under dietary influences. Incorporations of individual fatty acids into total lipids were calculated as increases in individual fatty acids as percentages of the increase in total fatty acids during growth, on a per animal basis. Relative incorporations (RI) so defined generated similar profiles for the...

Individual fish rhythm directs group feeding: a case study with sea bass juveniles (Dicentrarchus labrax) under self-demand feeding conditions ArchiMer
Millot, Sandie; Begout, Marie-laure.
The long term influence of individual biological rhythms on group feed demand behaviour was investigated in European sea bass (Dicentrarchus labrax) held under controlled environmental conditions with an on-demand feeding system. The experiment was realized over 219 days with 190 fish distributed on 4 tanks. Sea bass had a mean body mass comprised between 139 g to 183 g. The number of feed demand acts by each individual was calculated daily, and the population could thus be partitioned into three categories (high-, low-and zero-triggering fish). The duration of the period that an individual held high-triggering status could vary, but was 63 +/- 16 days on average. The transition period between two highest-triggering fish in one tank was on average 4 +/- 4...

Induction of ovulation and stimulation of spermation by LHRH or an LHRH analog associated or not with pimozide. Effects on some teleost fish. ArchiMer
Billard, R; Weil, C; Barnabe, G.
Effectiveness of LHRH and one of its analogs (LHRH-A = D-Ala super(6) ethylamide) on induction of ovulation and stimulation of spermiation is reviewed here. LHRH was also combined with pimozide, an anti-dopaminergic agent, to try to cancel dopamine inhibition of gonatotropin (GtH) release. LHRH-A alone (50 mu g/kg) did not initiate ovulation but only oocyte maturation. When pimozide (10 mg/kg) and LHRH-A (50 mu g/kg) were combined, gonadotropic hormone secretion was significantly increased, causing ovulation and stimulating spermiation. This combination advanced ovulation in trout. Trout egg quality was markedly altered by the pimozide treatment; it remains to be tested in carp. In sea bass, LHRH-A was much more effective than LHRH, and ovulation was...

Is it possible to influence European sea bass (Dicentrarchus labrax) juvenile metabolism by a nutritional conditioning during larval stage? ArchiMer
Vagner, Marie; Zambonino, Jose-luis; Robin, Jean; Person, Jeannine.
The purpose of this study was to check if it is possible to influence sea bass juvenile metabolism by a conditioning of larvae from day 6 post hatching to day 45 to a low or a high HUFA compound diet (LH, 0.8% EPA + DHA and HH, 2.2% EPA + DHA) when reared at 16 or 22 °C. Following a 3-month intermediate period (at 19 °C using a commercial diet), the adaptability of the 4 initial larval groups to a HUFA experimental deprived diet (0.5% EPA + DHA) were tested at 19 °C in a 60 day-experiment (d-151–211). The four experimental duplicated conditions were ex-LH16 and ex-HH16 for the 2 groups previously reared at 16 °C (initial weight, 7.3 ± 0.5 g) and ex-LH22 and ex-HH22 for the 2 groups previously reared at 22 °C (initial weight, 11.1 ± 0.5 g). Isolamento de Pasteurella spp. e Vibrio spp. em robalos (Dicentrarchus labrax): susceptibilidade a diferentes grupos de antibióticos Arq. Bras. Med. Vet. Zootec.
Saavedra,M.J.; Brito,R.D.; Sousa,M.; Alves,A.; Rema,P..
The most frequent infectious diseases that affect fish are those of bacterial origin. In European sea bass fish farms (Dicentrarchus labrax) are included Vibrio spp., Pasteurella piscicida and Myxobacter spp. In addition to these, it is also possible to find, although lesser frequently, other pathogenic agents such as Pseudomonas spp., Aeromonas spp., Staphylococcus epidermis, Streptococcus spp. and Enterobacter spp. The presence of these micro-organisms in fish farms contributes for a significant decrease in fish production and subsequent loss of profitability in these aquaculture units. The use of antibiotics may therefore be necessary as a prophylactic measure although their systematic utilization leads to the development of strains of antibiotic...

L'activite de deplacement et le reperage de la nourriture chez le bar juvenile captif ArchiMer
Anthouard, M; Paris, J.
Movement activity and localization of food were studied dynamically with the objective of establishing the capacities of behavioral adaptation of the young Dicentrarchus labrax faced with a novel situation. The role played by previous rearing conditions was demonstrated, the level of stimulation being a sensitive intervening factor (currents associated with the food). The data show the favourable effects of such treatment, and are in agreement with work carried out on higher vertebrates, which have brought to light the importance of early stimulation in behavioral ontogeny which respect to emotionality as well as capacities of adaptation in subjects reaching adulthood.

Le système immunitaire du bar Dicentrarchus labrax. Immunoglobulines, cellules productrices d'immunoglobulines, ontogenèse applications à une pathologie virale néonatale ArchiMer
Breuil, Gilles.
The sea bass, Dicentrarchus labrax, has been subjected to intensive aquaculture on the Mediterranean cost for ten years. Marine fish hatcheries are key factors in the aquaculture field, and face different pathologies, especially one of viral origin (nodavirus) that could challenge their feasibility. Consequently, we have tried to provide some data on larvae and spawning stocks. Firstly, we have studied the immunoglobulins, components of the specific humoral immunity, and their role as antibodies. On adult basses, these immunoglobulins are tetrameric molecules with high molecular weight (850 kD), made of heavy (H) - 80 kD and light (L) - 26 kD chains. In the eggs, simplest forms such as monomeric or dimeric are brought to the fore; they are located under...
